SUMMARY:Physics-Biology Interface seminar: Catherine Villard - Moyen Amphi\
 , Building 510\, Université Paris-Saclay Orsay - 19 Jan 24 11:00
DESCRIPTION:Fast deformation under flow and morphological recovery of cance
 r cells\nCatherine Villard (U. Paris-Cité)\nWarning: joint seminar with L
PS\, takes place at LPS\n\nThe cancer metastatic cascade includes a blood
circulation step for cells detached from the primary tumor. This stage inv
olves significant shear stress as well as large and fast deformations as t
he cells circulate through the microvasculature. The recovery dynamics aft
er deformation is pivotal to understand how a cell can pass through the mu
ltiple capillary constrictions encountered during a single hemodynamic cyc
le. \n\nI will present results on the recovery dynamics of circulating c
ells representative of different cancer phenotypes and their nuclei\, obta
ined thanks to a dedicated microfluidic system we have recently developed.
\n\nIn brief\, all cell types display an elastic recovery behavior followe
d by a slower viscoelastic regime\, while nuclei\, depending on the cell t
ype\, can strikingly recover purely elastically over a time scale of 20 ms
. The fast elastic cell recovery regime is under the control of cell contr
actility ensured by the integrity of the cell acto-myosin cortex while the
presence of a vimentin cage around the nuclei confers elastic properties
to this organelle.\n\nI will finally discuss how these behaviors can poss
ibly impact cell transit and survival into the blood circulation.
