SUMMARY:Physics-Biology interface seminar : Emmanuel Beaurepaire - Moyen Am
 phi\, Building 510\, Université Paris-Saclay Orsay - 29 Nov 12 11:00
DESCRIPTION:Nonlinear microscopy of developing tissues and embryos\nEmmanue
l Beaurepaire (Lab. for Optics and Biosciences\, École Polytechnique/CNRS
/INSERM)\nUnderstanding the dynamics at play during embryonic and tissue d
evelopment requires tissue-scale measurements with subcellular resolution.
Nonlinear microscopy is attractive for live studies because it provides t
issue 3D imaging and provides several contrast mechanisms. Intrinsic coher
ent signals such as second- and third-harmonic generation (SHG\, THG) prov
ide structural information and can be used in conjunction with fluorescenc
e (2PEF) imaging. SHG signals are specifically obtained from dense organiz
ed structures\, whereas THG signals are obtained from optical heterogeneit
ies and provide images of the tissue morphology. We will discuss these pri
nciples and applications such as imaging the microstructure of intact unst
ained corneas\, and reconstructing the early development of zebrafish embr
yos. Finally\, we will discuss ongoing efforts to improve the capabilities
of nonlinear microscopy for imaging developing tissues at advanced stages
\, such as wavefront control for aberration correction\, and simultaneous
excitation of multiple fluorescent proteins.\nReferences:\nSupatto et al\,
