 örster Resonance Energy Transfer (FRET) allows measuring the distance bet
 ween two spectrally distinct fluorophores\, in the 20-100 Å range. When m
 onitored at the single molecule level\, smFRET is useful in resolving subp
 opulations\, or observing conformational changes as a function of time wit
 hin single macromolecular complexes in vitro. I will describe here the met
 hodologies used to measure FRET accurately on single biomolecular complexe
 s\, freely diffusing in solution or immobilized on surfaces. I will then f
 ocus on our studies on the mechanism of prokaryotic transcription such as 
 antitermination control by the antiterminator LicT\, and Rho-induced trans
 cription termination.\n
